Differential effect of ethidium bromide and cytosine arabinoside on mitochondrial and nuclear DNA synthesis in HeLa cells.

چکیده
The effect of ethidium bromide (EB) on the synthesis of circular DNA of mammalian cells was studied by isopycnic centrifugation in a CsCl-EB solution. EB (0.1 —0.5 ^wg/ml) interferes with the synthesis of newly-formed circular D N A of HeLa cell mitochondria and causes degradation of the pre-existing circular DNA, as well. Under the same conditions, nuclear DNA synthesis was not inhibited. This effect was not reversible at a concentration of 0.5 /ug EB/ml or more. Cytosine arabinoside (ara-C) did not exhibit an effect similar to that of EB.

The Effect of Ethidium Bromide on Mitochondrial Dna Synthesis and Mitochondrial Dna Structure in Hela Cells
The synthesis of mitochondrial DNA (mDNA) in HeLa cells is selectively inhibited by relatively low concentrations of ethidium bromide. After exposure of cells to strongly inhibitory concentrations of the drug, the apparent superhelix density of mDNA is rapidly increased, as judged by its buoyant density in CsCl in the presence of ethidium bromide. Mitochondrial DNA synthesized in the presence o...
متن کاملEvidence that the synchronized production of new basal bodies is not associated with DNA synthesis in Stentor coeruleus.
Stentors were induced to produce synchronously thousands of new ciliated oral membranellar band basal bodies in less than 3 h. DNA synthesis does not accompany this process, as determined by [H]thymidine incorporation into isolated bands and by sensitivity to DNA synthesis inhibitors (mitomycin C, ethidium bromide, cytosine arabinoside and hydroxyurea). Yet DNA could be detected in the cortex a...
متن کاملSpecific Inhibition by Ethidium Bromide of Mitochondrial Dna Synthesis in Physarum Polycephalum
Eucaryotic cells contain two complete genetic systems, one based on nuclear DNA and the other on cytoplasmic DNA . The study of these systems has been greatly facilitated by the existence of specific inhibitors . One such inhibitor is the trypanocidal drug, ethidium bromide (Short, Peak, and Watkins, 1953) . Application of the drug to trypanosomes in vitro causes alterations in the structure of...
متن کاملCYTOGENETIC EFFECTS OF MAGNETIC AND RADIOFREQUENCY FIEL DS OF NMRI SYSTEMS ON THE FREQUENCY OF CHROMOSOMAL ABERRATIONS INHUMAN LYMPHOCYTES IN THE PRESENCE OR ABSENCE OF CYTOSINE ARABINOSIDE
Application of nuclear magnetic resonance imaging (NMRI) as a non-invasive and accurate imaging procedure has been widely used in recent years. Meanwhile, the biological effects of magnetic fields of several tesla (T) and high energy radiofrequency (RF) is not fully known yet. Because of controversy over this issue, the present research has been carried out in order to verify the effects of...
متن کاملIncrease in mitochondrial DNA quantity and impairment of oxidative phosphorylation in bovine fibroblast cells treated with ethidium bromide for 15 passages in culture.
Bovine fetal fibroblast cells were treated with ethidium bromide at a low concentration for 15 passages in culture to determine its effect on mitochondrial DNA copy number and on cell metabolism. Mitochondrial membrane potential and lactate production were estimated in order to characterize cell metabolism. In addition, mitochondrial DNA ND5 in proportion to a nuclear gene (luteinizing hormone ...
